Plant Induction by Anther Culture of Hot - pepper (Capsicum annuum L.)



Abstract
Five F©û hybrids of hot-pepper (Capsicum annuum L.) were used in the study for induction and growth of microspore derived embryos. Anthers in the mid-uninucleate to early bicellular stage were inoculated on the `C¢¥ medium (Dumas De Vaulx et al.¢¥s basic medium) supplemented with several growth regulators. The efficiency of anther culture was dependent on the genotype of donor plants and we obtained various culture efficiency from different genotypes. Induction of embryos from microspore was best on `C2¢¥ (0.1 §·/§¤ kinetin, 0.1 §·/§¤ 2, 4-D) and `C4¢¥ medium (0.1 §·/§¤ 2, 4-D, 0.1 §·/§¤ NAA). In experiment of heat treatment of anthers at 35 ¡É for 2, 5 and 8 days, 8 days treatment was more effective than 2 and 5 days. Growth condition of the donor plants had strong influence on the frequency of embryogenesis. Chromosome of thirty-six plantlets derived from anther were analyzed. The result was that 19 plantlets revealed spontaneous doubling chromosome (2n=2x=24) and the others 17 plantlets revealed haploid (2n=x=12).
